Retatrutide: research guide

Nordapep Research Team

Retatrutide is an investigational synthetic peptide that acts at three receptors at once: the GIP receptor, the GLP-1 receptor and the glucagon receptor. This guide covers what it is, what the research literature studies, how identity and purity are checked on a Certificate of Analysis, and how lyophilised material is handled. It is written for people working in laboratories, not for consumers.

Class
Investigational triple agonist (GIP, GLP-1 and glucagon receptors)
Structure
Synthetic peptide of roughly 40 amino acids, approx. 4.7 kDa
Supplied as
Lyophilised powder in vials; also pre-filled pen formats
Purity method
RP-HPLC, with intact-mass confirmation by mass spectrometry
Regulatory status
Investigational; not approved by any regulator (October 2026)

What retatrutide is

Retatrutide (developed by Eli Lilly under the code LY3437943) belongs to the incretin-related family of peptides. Earlier compounds in the family act on one receptor (GLP-1) or two (GLP-1 and GIP). Retatrutide adds the glucagon receptor, which is why it is described as a triple agonist.

It is a single synthetic peptide chain made by solid-phase peptide synthesis and purified by preparative chromatography, then freeze-dried (lyophilised) for stability.

What the research literature studies

Published work on triple-agonist peptides looks at receptor binding and signalling in cell-based assays, and at how combined GIP, GLP-1 and glucagon receptor activity differs from activity at fewer receptors. The mechanism is the subject of research interest; nothing on this page is a claim about effects in people or animals.

Retatrutide is also the subject of clinical trials run by its developer. Those trials concern the investigational medicine under regulatory oversight and are separate from laboratory research material.

Identity and purity: what a good CoA shows

For a peptide of this size, two tests carry most of the weight. RP-HPLC separates the target peptide from synthesis by-products and reports purity as the percentage of the total peak area. Mass spectrometry confirms the intact molecular mass, which is how you know the main peak is the right molecule and not a different sequence of similar polarity.

  • A batch reference that matches the label on your vial
  • The RP-HPLC chromatogram itself, not only a purity percentage
  • Observed intact mass alongside the theoretical mass
  • The testing laboratory's name and the date of analysis

Handling and storage

Lyophilised peptides are most stable cold, dry and out of light. Keep sealed vials in a freezer or refrigerator as directed on the label, avoid repeated warming and cooling, and let a vial reach room temperature before opening so condensation does not enter. Once a peptide is in solution its stability window is much shorter. See our guides on cold-chain handling and on reconstitution arithmetic for the general principles.

Regulatory status

Retatrutide is an investigational medicine and has not been approved by any regulator, including the MHRA, EMA and FDA. Material sold for laboratory research is not a medicine, has not been evaluated by any regulator, and must not be used in people or animals. Nordapep supplies it for in vitro research only.

Questions researchers ask

What does "triple agonist" mean?

It means a single molecule activates three receptors: the GIP receptor, the GLP-1 receptor and the glucagon receptor. Researchers compare this with compounds that act on only one or two of them.

How is the purity of retatrutide measured?

By reverse-phase HPLC, which separates the peptide from impurities, with mass spectrometry confirming the molecular mass. A good CoA shows the chromatogram and the observed mass, not just a percentage.

Is retatrutide an approved medicine?

No. It is an investigational medicine in clinical development and is not approved by any regulator. Nordapep's material is for in vitro laboratory research only.

Why does Nordapep sell both vials and pens?

Some laboratories prefer a lyophilised vial for in-lab reconstitution; others prefer a pre-filled format. Both carry a batch reference and Certificate of Analysis.

This guide is general reference material for laboratory research. It is not dosing, medical or clinical guidance, and the specifications quoted are commonly published figures: the Certificate of Analysis for your batch is what applies to your vial. All Nordapep products are supplied for in vitro laboratory research only. See the research-use disclaimer.
